PREPARATION OF MYCOPLASMA EXPERIENCE CONTRACT MEDIA
(APPLIES TO BOTH MYCOPLASMA AND UREAPLASMA MEDIA)
| SOLID MEDIA | ||
| Mycoplasma Agar | 325 ml. | 65 ml. |
| Supplement for Mycoplasma Agar | 175 ml. | 35 ml. |
| Volumes Prepared:- | 500 ml. | 100 ml. |
| Ureaplasma Agar | 300 ml. | 60 ml. |
| Supplement for Ureaplasma Agar | 200 ml. | 40 ml. |
| Volumes Prepared:- | 500 ml. | 100 ml. |
Storage temperatures - Agar (4°) Supplement (-20°C)
| 1. | Melt the agar component in a boiling waterbath, allow to cool slightly before placing in a 50°C waterbath. |
| 2. | Allow the frozen supplement to thaw at 36°C. Mix gently when completely thawed. |
| 3. | Place the supplements in the 50°C waterbath with the agar. |
| Note: | For the agar temperature to fall sufficiently after placing in the 50°C waterbath, allow ~30 minutes for large volumes and ~15 minutes for small volumes. |
| For the supplement temperature to rise sufficiently after placing in the 50°C waterbath, allow ~10 minutes for large volumes and ~5 minutes for small volumes. | |
| 4. | Mix the agar and supplement thoroughly whilst avoiding bubbles and distribute 8 ml. amounts into 60 mm petri-dishes. |
| 5. | When set, dry the plates inverted at 36°C for 30 minutes. Store at 4°C. |
LIQUID MEDIA
| Mycoplasma and Ureaplasma Media | 500 ml. | 100 ml. |
| 1. | Allow the frozen media to thaw at 36°C. Mix gently when completely thawed. |
| 2. | Distribute into air-tight containers. Store at 4°C. |
| (Solid and Liquid media may be stored at 4°C for at least 14 days). |
